Autoradiography

Satisfactory autoradiographs are readily obtained from wet, unfixed polyacrylamide gels containing 125I, 32P, or high-specific-activity 35S-labeled proteins. The entire preparation of proteins can be labeled, or the sample can be spiked with a small amount of labeled protein. If a small amount of purified or nearly purified antigen is available, it can often be iodinated and used to spike a small sample before running on the gel. Because of the high specific activities that are possible using iodination, this variation often allows for the easy detection and localization of antigens.
