Preparing Myeloma Cell Feeder Layer Plates

Myeloma feeder cells are popular because they are readily available and easy to grow in sufficient numbers. They provide essential growth factors that help the newly formed hybridomas survive in low-density cell culture until hybridoma colonies can grow out and produce their own growth factors. Because they have a mutation in the HPRT gene needed for the nucleotide salvage pathway, they will not survive in the fusion selection medium.
