Antibody Capture on Nitrocellulose Membrane: Dot Blot

A dot blot is an appropriate screening procedure when the antigen is a protein that is available in purified form. The antigen is bound directly to a nitrocellulose sheet and incubated with hybridoma tissue culture supernatant. A dot blot is widely used to determine the productivity of a given hybridoma, and this is described here. This assay can also be used to screen a fusion or subclone plate for productive hybridoma clones. The protocol described here is adapted from Sharon et al. (1979), Glenney et al. (1982), Hawkes et al. (1982), Herbrink et al. (1982), Huet et al. (1982), Yen and Webster (1982), and Vera-Cabrera et al. (1999). For a review, consult Hawkes (1986).
